I'm thinking of playing a 4-4-2 with an assymetrical wingback.
Will my formation experience still be "excellent" even though I have never tried this before?

Yes.
And it's trainable as long as you don't start 4 players at Central Defender or Inner Midfielder.
4-4-2 is always excellent, IIRC.

FYI, for formation experience only the numbers in each area matter, not how you get there. (i.e., a 3-5-2 with two wingbacks is counted the same as a 3-5-2 with three central defenders.)
