I've heard in a number of places that there are more bad players at Partypoker but I haven't played on the real money tables at Pokerstars so I can't compare it myself.
if you want a challenge.... PokerStars
if you want to win money.... PartyPoker

I would opt for the Party Poker network just for the sheer number of players that feed into its card rooms through Party, Empire, PokerNow, MultiPoker, Intertops and Eurobet. If you play a tight-aggressive style, you are bound to do well there.
You would also do well to look into the Crypto network (Interpoker, Caribbean Sun, PokerPlex, Wm Hill, TotalBet, et al) as there are a large number of fish swimming around in their network and you can get over $300 in loyalty bonuses combined from their sites each month.
In the interest of fairness, I have not played PokerStars, but my little brother swears by it - from a gaming perspective, not a "making money" perspective. It seems to be universally accepted as the best interface/software out there. However, the others aren't so much worse as to exclude them from consideration.

I'm hesitant to try Party Poker just because I like the PokerStars software so much, but I've heard the same that's been listed - Party Poker is where you can win, PokerStars has the best software

isnt winning the point of playing especially with real money :) , if it easier to win at partypoker than at pokerstars.

If you want a challenge, then challenge yourself to extract the maximum amount of money from the fishies at Party ;)
Poker stars is actually pretty easy to win at the lower levels (they don't even have anything lower than .5/1 at Party). I choose Stars because it has lower limits which I like to play (less risk) and also the players aren't as wild (and still plenty beatable at the lower limits, again) so you can still make money with lower variance (albeit a lower win rate, but I will take that with less variance, personally).

haven't invested any $ yet. still playing the free tables...which are way too easy
