Absolutely Hank knows.
Once he read that note it all came crashing down on him. The "gambling addiction", the new cars, the car wash... Hank's a smart guy and obsessed with this, things have just clicked for him. It's how he reacts is the big question. He's tied up in this. Walt's drug money allowed him to walk again.
