I'd have to say any secondary position makes the most impact. You rely on your corners to cover the pass, and your safeties provide run support and also help out with pass coverage. Plus, most of the time you probably don't even take control of a CB when you play the game, thus making this position more important because you rely on the AI.

Linebacker. A dominant LB is the core of your defense and is CRUCIAL in stopping the run.

Got to Be DT and DE

He can affect 2 things.

Passrush and stopping the run and less time for QB to throw the ball which helps your D allowing your team to get better field position and making it easier for your offense to score.

linebacker and o line, if you have no oline no matter who your qb is they get killed, and your running game most likely will suck.
