I am playing dynasty mode with Notre Dame and I recently received an invite to the MAC. I am wondering if I accepted it , later on could I join one of the bigger conferences like the Big 10/Big East/etc...?

You should be able to. I've gone with a team from the Sun Belt to C-USA and then to the ACC.
