If I change sliders in week one, do they take effect immediately and any games started after the change will have the tweaked sliders? or do they not come into play until week 2?

Yes, they will kick-in immediately.

Now, some people believe that they need another week to really "take root", but I have seen nothing in my experience that either proves or disproves that.
