MOFO = Middle Of the Field Open.
MOFC = Middle Of the Field Close.
How I have this interpreted is this indicates how the safeties/DB's are lined up deep. For example, Cover 2 defenses have 2 deep safeties, one to each side of the field, leaving the middle open. In a Cover 3, for example, you have 2 DB's deep on either side of the field, plus a deep middle defender, so you're closed on the middles.
To beat a MOFO defense you want to use routes that go inside, like slants and In routes,
To beat MOFC you want to lean towards outside passes such as Out routes and Floods.
