Just wondering, if I preorder on Xbox One digitally, will the game be playable at 12am EST Friday morning (Thursday night) or 3am, meaning 12 PST?

This is incorrect. A little known tip for Xbox users is that you can change your region to somewhere else and actually access that region's marketplace. So what myself and many other people do is change it to Hong Kong. They are 12 hours ahead of the US. Then once it hits 12am in Hong Kong, people who bought it from their marketplace will have access to it by noon Thursday.

Edit: The price says $468 but that's not the US dollar. It's the Hong Kong dollar. $468 HK turns out to by $60.39.

Wish I knew this before I made my purchase, but to be clear we would have to literally buy it under the Hong Kong profile, or could we just change it on Thursday to Hong Kong to get it to play early even if we bought the game under a US account.

You must buy it from that marketplace. Simply changing your region to Hong Kong after you've already bought it from the US store will not unlock the game.

Can you change it back right after the transaction, or do you need keep it set on Hong Kong until the game unlocks?

So you can still preorder off the marketplace and be able to play Thursday night ? Like there wasn't a cutoff date or anything.

Yes, you can change it right back. You do not need to keep it set on Hong Kong. I said Hong Kong since their dollar currency exchange will still be around $60 US. You could buy it on New Zealand's but their exchange is a little higher, you would end up paying $70.
