Do you hit it as soon as your rb gets the ball or do you wait till you get through the line or to the outside, depending on the play called?

It depends on the play type. If it's a run up the middle and I only need a yard or so I hit it as soon as I get the ball but if it's a regular run up the middle and/or a bounce run outside then I wait until after i'm through the hole and I need the boost.

When running my 1st move is a juke or stiff arm. Spin if necessary. I don't start sprinting until I have a more open field. The only time I will sprint right when handed the ball is on a draw play. Especially one which the defenders drop back in coverage and don't blitz. That's always good for a decent gain.
