4-3 seems to be best against the CPU. You can take a safety and run a cover 2 man 90% of the time and be pretty stout.
People seem to read the 4-3 pretty easily though because it is more common. I like to run the 4-2-5 for that reason - humans have a really hard time figuring out what you're going to do out of that formation (or 3-3-5) in comparison. They don't see it very often and the extra safety can do a lot. 4-6 is also a favorite of mine when you're playing someone who is run first.

I think your right as far as 4-3 being the best against the cpu (unless your loaded with real fast, good tackling LB's), although I usually control the MLB. I find I screw up the coverages too much on deeper passes when I control the safety.
