The only significant change is locomotion. That makes it boil down to one question:

Do you like holding R2 to control running or do you want the computer to dictate that?

Madden 10 - Holding R2
Madden 11 - Automatic

I don't have any preference so I went for the cheaper title. Although I really don't like how Madden 10 deals with tired running backs. If you hold the R2 for more than 5-6 seconds, your player starts limping from "exhaustion" - even if it's still the 1st quarter and he's a 92+ OVR. It's the weirdest animation ever and has stopped me from so many easy touchdowns. Madden 11 probably fixes that by automatically letting go of the sprint button when your RB is tired, but that just doesn't seem plausible to me. When you're breaking away (10 yards ahead of everyone), the adrenaline should kick in and you should be sprinting regardless of any tiredness. Or if you're going to make the running back tired, at least don't make the "tired" animation look so atrociously glitchy.
