apart from editing a player manually, will players be able to develop/decline signature skills over time automatically?

Players will gain AND lose signature skills over time in association mode. Examples:

Anthony Davis gained Defensive Anchor after year 1

Dwayne Wade lost Finisher and Acrobat as he got older.

In my career DWade lost Finisher and got deadeye. Prolly because he mostly was shooting high percentage jumpers with me at point

Id say they are based on Ratings and possibly even player type.

Be nice if 2k actually gave some clarity on some things once in a while!

I'd say they've been programmed into the roster by the 2k Insider to update over seasons progressively and that regenerated players don't gain new sig skills over time.

Regenerated players? If you mean generated players, they also gain signature skills over time.

They also gain / loss abilities in Myplayer mode as well.
