Are we forced with our number 1 starters for online play?

Also,are we allowed to sub in starters for relief?

Well the answer to your first question is this.
You can adjust your lineups and starting pitcher before the game.

I am pretty sure ASB_Man stated that you can bring in starters during the game however.
