Here are the standings (I didn't include "OR" voting because the thread title specifically says ONE game, and let me note that GTA would have had like 4 or 5 votes if it wasn't pushed back )

1. 14 ... NBA 2K8
2. 10 ... Mass Effect

4. 6 ... Guitar Hero III
5. 5 ... NHL 08
6. 4 ... Call of Duty 4

Wow, I'm surprised GH III has 6 votes and Rock Band has none (or just one). I'm a huge GH fan, but Rock Band looks and sounds insane. Plus, Harmonix made GH what it is today, I credit them with the series' success and fully believe Rock Band will follow suit.

Now that I've said all that, my ONE game is Bioshock. But if I could only get one game this year (in reality) I do not know what I would do with myself because there are just too many great titles coming out.

I think right now rock band is to much of a question mark price wise for people to pick it

I can't decide between Mass Effect and NBA 2K8. It would have been GTA though.

I'm gonna go NHL 08, every year I play the hell out of that game and all the previews this year have been amazing. IGN is already saying it's probably going to be the sports game of the year. Don't think I'll have a PS3 on release date but will within a week or two. This is the game that's making me jump to next gen, COD4, and Assassins Creed which are probably my next most anticipated games are just bonuses.
