This is kind of a difficult genre to pick from because it encompasses so many major sub-genres. You have JRPG's like Final Fantasy, Action RPG's like Skyrim, Tactical RPG's like Fire Emblem, etc...So maybe it would be better to break it out by those sub-genre's
JRPG - Xenosage 1.
Really it's the entire Xenosaga trilogy that deserves to be mentioned as they all go together, but I'll list the first one since it is what got me hooked into the saga. For me, JRPG's are all about the story and its characters, and no RPG (no other game period for that matter) has ever delivered the epic story and memorable characters that Xenosaga did. At times the game is like watching a movie with its very lengthy cutscenes, but I actually loved that about the game because it was so story driven. Those long scenes just really pull you into the universe and give you a feel for what is going on.
Action RPG - Mass Effect 2.
I probably would have picked Mass Effect 1 if it had the same gameplay improvements that the sequel did, but I really can't be upset picking ME2 either. To me the Mass Effect series has the deepest and most compelling story of any game outside of Xenosaga, and the 2nd installment in the series really upped the stakes. Also, few games really let you make a character your own in the way the ME series does with all the story impacting choices you make throughout.
Tactical RPG - Disgaea: Hour of Darkness
I don't play a ton of these, but I'll list the one that I have definitely played the most. The learning curve for the combat is pretty steep if you really want to master the game, but it's fun leveling up your characters to maximize their potential. Also, Prinnies.
Some honorable mentions I'd like to add, but I'll wait to see what others say.
