DOWNLOAD Franchise file with: 25 Man MLB Set, Opening Day DL Assignments, and 40 Man Rosters.
Updated 5/29: Thanks to HustlinOwl who ran though and cleaned up some things I missed!

This is a 30 team Franchise File at the end of Spring Training (April 3rd to be exact) based off Kinghts (amazing) V2 release, without impacting player Options or the Rule 5 draft, that includes the following:
All accurate DL assignments as of April 7th. This includes guys that are on the 60 and 15 DL as of the 7th.
Accurate 25 Man MLB Rosters (with a few exceptions, see below)
Accurate opening day starting rotations and lineups (that will stick when you click 'advance to regular season').
All 30 team 40 MAN Rosters as of opening day. (as close as possible, some are still short a player or two)
To control the team you want for the franchise, follow these steps:
Load the file.
Go to 'Options' menu
Go to 'Profile'
Click 'Profile Associations'
Use the right stick to assign your profile to the team you want to control.
NEXT, GO TO SCHEDULE AND ON APRIL 4th press 'X', and click advance to Regular Season.
THEN, If you'd like INJURIES to happen during your season, (other than the real life opening day ones I've set) you MUST got to OPTIONS, and turn them back to ON. As well as any other 30 team options you deem appropriate.
If you follow real life day to day injuries and will assign them accordingly, keep as Manual.
TRADES are also currently set to Manual. You'll need to set to Auto have the AI trade and propose trades. Remember, you have full veto power with 30 team control.
Notes:
Oakland had too many DL assignments to start the season (giggle), so Coco Crisp is on the 25 man.
Due to circumstances beyond control, the following rosters are actually 24 men on April 4th. (the CPU will add a player for you, perfection is hard to achieve!) Florida, Chicago (NL), Colorado, TB, and Detroit.
The following have 26 men on the roster. The CPU will designate an additional one to the minors. Again, perfection...NY (NL), Cincinnati, Arizona, Boston, and Oakland.
You'll notice that many of the AAA Rosters are heavy several guys before you advance to the regular season. Unfortunately, there is no way around this. However, the CPU does a surprisingly good job of sorting the minors out when you click advance to regular season. Ultimately, you can move men between systems without penalty if you choose, so no biggie. Since you have 30 team control, you can go through and mop these up if you like.

Here's how I run Global settings on my Franchise (remember, Manual doesn't mean you HAVE to do these things, it just means the CPU will prompt you. If it's something minor like a AA injury, just let the CPU fix it. However, it's nice to know when Prince Fielder gets hurt so you can make the appropriate roster moves, lineup changes for the CPU.)
Also, when they ask, "Auto-Fix" always choose, PLACE ON BENCH, then 'circle button' out, and go fix. (I have a little notepad handy to remember team/name if their are more than one...) If you don't do it this way, batting orders and rotations will fall apart on you.
Injuries: ON
Lineups/Rotations: Manual
Roster Moves: Auto
40-Man Roster: Auto
Waivers: Auto
Injury Management: Manual
Trades: Auto (you still have Veto power)
All Operational issues to: Auto
