It says in game too. 0 is what you want if you want a bigger gap between fast runners and slow runners.

This user is correct, thats how any sort of threshold works. You set the point where you want anything below it to be equal to that point. Basically if set at zero then a 76 will outrun a 75, if set at 100 then everyone is basically equal.

And to be a little more detailed, according to what the developers said on the EA Sports Inside Blog site, they said that in past Maddens, the fastest players on the game were running about a 4.2 40-yard dash time, while the slowest were over 7.0 seconds. So they made adjustments such that everyone supposedly matches their in-game actual speed, when playing All-Pro or All-Madden with the Speed Differential set to '50'. But for Rookie and Pro, the Speed Differential at '50' resembles previous Maddens, where the difference between faster and slower players is greater, so that slow players are actually running more slowly than they would in the real NFL.

But they said they put the slider in there in case people who play on Rookie or Pro want the players to be more realistic or people who play on All-Pro/Madden to have the faster players feel much faster as in previous Maddens.

The problem I have is I don't know what the 'realistic' setting is for Custom Player Settings. By inspecting Practice Mode replays, I believe '50' is it, but I am not 100% positive.
