The most basic indicator: If they're in zone and have two safeties deep, it's Cover 2, if they have only one deep presnap, it's Cover 3. Not always completely correct, of course, but it's the easiest way to get a hint what kind of zone they're running.

Actually, watch the safeties and the corners. If the corners and one safety drop back it's cover 3.

With the exception of cover 3 press... the corners play up on the receivers, but one safety will still drop down.

To the OP the only real way to test cover 2 if you suspect it is, is to motion an outside receiver to the opposite side of the field, if the corner responsible for that receiver follows him it's man (cover 2 and man/man under look similar) if he doesn't then it's cover 2 or some other form of zone (cover 4 can look like cover 2 because both safeties stay high but the corners play off). Against a human opponent it's a little more difficult if they show blitz or manually move a safety so be careful.

When the FS rotates over to the center of the field it is most often cover-3.

But there is also a "Cover-3 Bluff" play.

With Cover-2 zone, you can watch the CBs once the ball is snapped. IF they press and jam the reciever but "stop chasing" and pass him off to the safety behind them its a cover-2 zone ussually.

With Cover 2 run corner routes, or flood routes, or smash routes.

The cover 3 bluff play that you are referring to is a cover 3 coverage. The bluff potion of the play is that it fakes an overload blitz on the left side of the line. Same principle that is behind the Sugar Cover 3 Bluff except the Sugar bluff is bluffing A gap pressure. The plays that disguise coverages as something else are the roll plays that give a presnap look of one thing before rolling of a safety or a pair of safeties to form the cover 2/cover 3 shell.
