There are so many "best moments," it just depends on what sports you like, what teams you like, etc.
As far as most significant moments, that might be a little easier to compile. My top two most significant moments would be 1. Jackie Robinson- no explanation needed 2. Jesse Owens, 1936 Berlin Olympics- dominated like no one had ever done before, and in Hitlers backyard."You can not ensure success, but you can deserve it." - John Quincy Adams

Of course, Liston tried cheating by putting a foreign substance on his gloves to burn and blind Ali. At one point Ali told his trainers to stop the fight but they kept going. Is the PK Sam catch the one where he ran out of the end zone a dog nearly bit him? That might have been someone else, but I distinctly remember someone catching a pass that game and a police dog almost biting him.
